PCR - The expanded post
Ok a lot of people are wondering why I said PCR is prone to false positives and bad results so I'll try to explain it here.

For PCR to work it needs something to stick to the beginning and end of the DNA strand that is to be amplified. Now this thing are called primers and these primers are designed by being complements of the actual part it is supposed to stick to. Think of primers as having a shape this will fit a pattern and the pattern is supposed to be the DNA end. Sometimes if enough of the shape is close enough the primers can stick to another DNA which closely resembles the primer's original shape. So this would cause what is known as non-specific PCR amplification giving you amplification of some other part of the DNA than you originally intended. This problem can be solved be redesigning the primer to be more specific (making the shape more complex and such) or by altering the conditions to make it harder for wrong pairings to occur (increasing PCR temperature).

A PCR result does not confirm the identity of a gene amplified .. lets say you have a DNA and you design a primer to attach to certain parts of the DNA to create amplifications of a 2.2 kb (kilo base pairs , measurement unit for DNA) sized DNA and you do the PCR .. then you get a 2.2 kb sized band .. it doesnt mean it is the original band you targeted. Why ? Lets look at an analogy ....

Imagine the gene being amplified as a story book , Cinderella for example . Now the book Cinderella has a beginning and an ending. We design primers to stick to the beginning and end . We design it to recognise " Once apon a time " as the beginning and " they all lived happily ever after" as the ending. Now that we have our primers we do PCR on the book (DNA) we get results and the size that is amplified matches the original book (200 pages) ... this doesnt proof that the book is Cinderella since there are many fairy tale stories which begin and end the same way. The better way to know would be to read the whole book and compare it and for that other more reliable methods are used.

PCR of DNA for use in court cases has been modified so that it can be used to compare two DNA samples and say if they are the same or not so no problems there but remember to have extra proof of a DNA's identity (like sequencing the gene) before saying that the PCR DNA is your DNA of interest. Normally we call PCR result as putatative results since it has to be confirmed with other methods ... (yes sometimes we go on faith that if I was supposed to PCR Cinderella and I get an amplified DNA which matches the size that I predicted it should be Cinderella but that is for non-critical applications only)

Ok , hope this cleared up some questions. I can give the full 10 min leture when we meet up for DotA next if you guys want .. I can talk about DNA all day long if I have to but dont bother since normally people ask and get bored half way through :)

PCR
PCR stands for Polymerase Chain Reaction .. if you throw away the fancy words it refers to a method which will amplify small amounts of DNA into larger quantity. The method works like a chain reaction where you put in a chemical which can syntesize new DNA based on the original sample and the reaction will go faster and faster (since there will be more DNA everytime the chemical makes a new DNA strand .. 2 -> 4 -> 8 -> 16 and so on).

PCR is important in any field that involves manipulation of DNA. In forensics (you see this in shows like CSI ) we can identify a person based on DNA from a single strand of hair / skin cells - they use PCR to do this. Small amount of DNA becomes large amounts of DNA and can be generated as much as is needed. PCR unfortunately is contamination prone and is also knows to give false positive results. One popular running joke in the lab is that in Malaysian court cases when DNA evidence is produced, 3/5 times the murderer is the lab technician :p

If anyone is actually interested i can write an article on this but I think this much is enough for the general layman.

Ah yes, to satisfy your curiosity .. I am not dead. :)

Instead, in between my thesis writing and Anarchy Online i've not had a minute's free time as any free time scrounged up would automatically be entered as "Sleep Time" which is more or less the only thing keeping me sane (not crazy would be more appropriate .. if there were borders indicating Sane and Not Quite Sane ... I've dodged the border patrol and am now hiding in the local suburb sewer eating dead rats or leets for a week ) Without further ado, here is the big post that i figure that i owe you people for keeping the blog alive for so long without me.


I can talk about my thesis writing and rant about it but there is only so much you can rant about how genes are organised in the cell :P so lets move on to the other world that I live on .. the world of Rubi-Ka. A highly technologically advanced Notum mining world which is torn apart by battles between the Omni-Tek company which legally owns the planet and the Joint Clans who believe the planet belongs to the people.

My journey to Rubi-Ka was done through the eyes of Serra Stormshade .. a young idealistic Opifex (think Sci-fi version of elf) who believes that one person can make the difference in the struggle for survival and freedom. Faced with misfortune the shuttle that she took down to the planet crashed but fortunately she survived ... wading to the shore with tired arms and a heavy heart she realised that she is lucky to be alive but her death would have had no meaning in the contant battle going on in Rubi-Ka. Deciding that she can make a difference , she joined the Joint Clan Alliance as a freedomfighter. This young Agent .. regarded as a terrorist by Omni-Tek, for that is what one man's freedom fighter is to another, made quick friends with the Martial Artist Itame, the Enforcer Janorath and the Adventurer February5. I would chronicle their tale here but I felt that, to do justice to their story they should have to do it themselves.

The first picture is of Storm looking at the sinking shuttle which very nearly became her cold impersonal tomb. As a new fledgling Agent she had to fight many scary and wonderous creatures ...




Fortunately she wasn't left to fend for herself in her struggle. In the city of West Athens where she arrived after joining the clans she met up with a kind soul called Good Boss .. concerned with the lack of balance between the people who are both new and those who are very experienced in the world of Rubi-Ka he decided to fund and start an organisation for clan newbies where they can help each other. This was Good Spirits. The motto of the org is "Froobs can win !" and it seeks to become one of the titanic power in Rubi-Ka but instead of using a harsh military discipline or soulless bureocracy , the org seeks to win by relying on the bonds of friendship. Will the org succeed ? We think so.


The first time Storm saw this she thought that the end of the world was coming .. it reminded her so much of an old old movie she saw on the shuttle trip to Rubi-Ka called Independence Day .. together with Itame they kicked up a big fuss and tried to warn the people in the city , only to be laughed at since giant battleships like this were very common .. especially with the alien threat to the planet.




Storm has met many interesting people during her adventures .. like this guy Crazypants , who walks around Old Athens in a manbra/bikini set. He has achieved something akin to notoriority in the city and most people know him by name or at least by description.



Well life in Rubi-Ka is quite fun and Storm does get to indulge in her favourite hobby of photography. Fortunately for me, there are some real life females who are daring enough to model for Storm. Well, always wanted to be a photographer .. maybe through Storm's eyes I get to live it up.


As you can see , people in Rubi-Ka know how to have fun ....




... and even get a little bit kinky doing it at times. But hey, it's just in the name of fun and there is no real harm in doing so. After all , to deny our very own human impulses is to deny our very nature itself.




So what are you waiting for ? Come on in the notum is just fine.

Not half as bad....
I nearly bought xbox360 on monday, but sadly no stock. dammit. well, i guess i just wait for the xbox 360 elite to be here. xbox360 is well known for it's over heating problems and once 3 red lights in seen, then send to hospital also cannot cure liao. unless get the unmodified one, which i m planning to get. I can borrow games from my fren and if there is offer in the net, i can grab the game for a cheap price around rm80 or so. Overheating is still the main issue ler. sad....... wii no game lar, and mostly chibified character one, not so my style. PS3 at the moment no games also, the best time to get ps3 is next year, and probably i will get 360 first for this year and ps3 for next year.

Some of the games for 360 is really stunning. Assasin Creed is one for certain, the games is somewhat akin to GTA and it has great graphics and actions (wading through the crowds and push people around). another one soon to be released is Lost Oddesey (spelling?) by Hironobu Sakaguchi (familiar name? the father of final fantasy) whom is working for Mistwalker now, company owned my microsoft. No ff game though. so we can expect good games from this company hopefully. i for one like 99 nights and i think it's an awesome game. oh, and since u all like FPS like CS, War of Gear is definitely the game for all of u, try play online. The game has one of the top notch quaility graphic i have ever seen. since seeing ps3 and xbox360 games, i think ff12 and kh2 (used to be super beautiful graphics to me) graphic now is just a small fragments of what i have seen in the new gen consoles.

The price for xbox360 limited set in my place here is rm1700 comes with 2 original games. i asked around at kl there and there price is rm1800+, i dunno y so expensive and the game for 360 (pirated of coz) is around rm18-20, which is quite taxing compared to rm15 here. any explanation or is ther any cheaper place around?

with xbox360, we can play final fantasy 11 ler. u guys getting it? magnus, heard u planning to get one ^^ from the chatbox. probably i will just wait until early next month then i c what to do lar. ps3 is definitely out for me in this year and wii i only like dragon quest. xbox360 is the best choice for now.

28 Weeks Later....the Blog is still dead....
Well, I ran out of days for my free World of Warcraft account and now have to cough up roughly RM50-60 per month to play on top of another RM155 to buy the expansion pack (which does give me a free month). Still I really think this is a game I should play, but I'm hesitant since we're playing Anarchy Online and my brother will be coming back at the end of the month (i.e. fre weekends in K.L. will be infrequent). This will leave me with some playtime on weekdays before my roomate gets home.

What do you think? Should I play WoW? The upside is I have cousins in WoW who will help me out, the downside is that its going to come down to a choice between WoW and AO. Its funny, I hated AO at first, then I enjoyed it, then I hated it again, and now its fun. Its the online communitee I guess, the people there are fun and make it worth playing, I wasn't in WoW long enough to feel that.

I guess I'll postpone WoW for now, after all Adole said .hack//G.U. Volume 2 is out, which means I'm going to buy it today and play like mad over the weekend. Yup not going to Seremban since there's no point anymore, DotA is a dead practice and Seremban in itself is boring so its still only after the end of the month for me.

I also managed to focus and finish Digital Devil Saga 2 on Wednesday evening since the AO servers were down for maintainance. Definately worth it, finished the final dungeon in one playthrough, but it was insanely annoying, especially the 2 bosses before the final boss, Ravana and Megananda, I wanted to give up, but I made it through and finished the game. I was so happy that Heat joined (even though I knew he would since I was following the walkthrough to make sure he joined). my only question is what the hell is the word?!?!? That 'one word was all we needed to know' What's the damn word!?!?!?! Adole? Do you know?

Adole any idea where I can get the DDS2/1 soundtracks? I don't mind getting CDs, bluelaguna doesn't have the tunes and no luck with limewire or torrents. Any advice?

I'm meeting my friend from Sabah after work today for coffee ad then spending the rest of the weekend gaming. I have another old A-levels mate coming down from London in June as well. So while it seems that I am whiling away the time gaming, I'm just doing this since June will be an overload of activity, more so if I have to make a trip to the States if my fiancee can't come down here (stupid malaysian visa requirements).

Well that's it for now. I probably won't post anymore after this. Will comment from time to time or shout out.

I got a copy of free dvd of the client 2 days ago (i dun have time to blog, sorry) and i have instlaled and patched the games due today. So today, the open beta is officially..........erm.....open. but Fuck, i m leaving for kl tonite!!!!! ARGGGGG/.but i m taking a half day leave lar, after packing and all, i m going to try it out!!!!

The game is kinda unique and it's different from any mmorpg. in this game, u can create up to 3 characters. and u control all of them. so, the party is so called squad now, meaning 2 people will have 6 characters. just like on the www said, u dun have to beg for healer to join ur party anymore, just create one for goodness sake.

I think i will be playing wizard (offensive and defensive magics), elementalist (pure offensive magics) and a warrior (my meat shield, brawn and no brain one). or mayb i will create a scout (healer and a bit of rouge like). I duno yet, i will test it out. i m on 1 day leave on monday!!! yea, then i can really try game then !!! will update soon enuf.

Return from the red light district
So I finally arrived in shitty old Malaysia and got home at 12:30am wednesday morning only to come into work at 9:30am and find that my fucking computer wasn't working, and it took the fuckers till 4pm to fix it after much shouting and cursing from me (mind you the network was screwed since monday, but I had to come in on Wednesday and kick up a fuss to get them to fix it). Why the fuck can a highschool dropout in Seremban keep and maintain a network at the cybercafe, but the so-called educated degree holding monkeys here can't even keep an open connection to the fucking server. DUMB FUCKS!!!!! Hope they all get cancer, HIV/AIDS and other painful wasting diseases....you think I kid but I'm really not.

I was on a business trip to Bangkok and didn't really do much. Arrived on Monday afternoon, had meeting till 5, decided to stay at the hotel for dinner since half of the people going out for seafood dinner were smokers and we would have wonderful malaysian food.....in Thailand. So I stayed at the hotel and ate like mad, at the RM100 buffet, cos hey its free for me. I took some pics that I will update soon once I get home (not many since my phone died from the dreaded no-signal battery drain).

I had it all, sushi, clams, oysters, lobsters, salmon, bass, tuna, cod, lamb, beef, chicken, duck, squid, octopus, and topped it all off with peach cheese cake and tiramisu. Slept after watching to TV (that Adam Sandler movie about playing football in prison). Woke up and had a buffet breakfast of bacon, ham, steak, bacons, pancakes, bacon, blueberry french toast, bacon, coffee, bacon. HEY there was bacon there and by fuck I WILL EAT BACON IF IT CROSSES MY PATH!!!!!!!!!!!!!!!!!!!

After meeting on Tuesday we went to the airport where I had a nice dose of glass noodle salad and green curry, which promptly gave me food poisoning and I spent my entire turbulence ridden flight back trying not to throw up, which almost happended when they tried to serve me chicken satay and ketupat on the plane itself. Reached home, logged online to email my fiancee that I was home and passed out.

Thus concludes the orgy of food that was my Bangkok trip. Sorry, no ladyboys

To cuss or not to cuss......
Lately i got told off by my friend for using a tamil cuss word(nearly in ranking with fuck) in front of her.She said she gets offended everytime i use the word even if i dont direct it at her,saying that it shows that i dont respect her.
To tell the truth i do use the cuss words a bit frequently,not directly at sumone but normally in replacing the word DAMN,and most of the people around me dont seem to bother. I havent got any angry stares from them either. But now after this incident im starting to wonder how many other people have i offended.
If you notice,there are many people who rampantly use the cuss words amongst friends(in discussions,on blogs,in sms,in a party) and no one really cares nowadays....rite??im not sure......I mean in a meeting or any public place these words are forbidden (i know) but those places i mentioned...wats your take on it?

Its a habit of mine and i want to decide wether i should put stop to it depending on what u guys think

And rate yourself on how good looking you think you are, on a scale of 1(so hideous that your mother tried to shove you back up her birth canal after you were born) to 10(so good looking that your mother tried to shove you back up her birth canal after you were born).

I haven't a clue how tall I really am, the last time I was properly measured was a few years ago, and I've grown a lot since. If I were to guess now I'd say 5'8" or a little more. 5'9"-ish. I think. Anyone have those scale thingy majiggys?!?! :S

I'm also severely underweight, but its NOT MY FAULT Y'ALL! I have an insane metabolism. I used to take Appeton weight Gain, but it never worked. So my mother gave up.

Welcome..to the World....of Warcraft


Oh hell yes. After 3 days of downloading 500MB of patched (which explains my lack of presence in AO) and another 500MB of patches that Sai burned for me (thanks man I owe you), a whole bloody Gigabyte of patching!!!!!!, I can play World of Warcraft.

The reports are. Not laggy at all, granted that the starting undead village didn't have more than 6 players that I could see. I'm hoping that there will be no problems. Going to try levelling my Undead Warrior to Level 10 and head out to do a final lag test before I go out and buy "WoW: The Burning Crusade". What can I say, I gotta make me a blood elf Paladdin (the dude in the picture). I wanted Undead, but they look kinda stupid, although mine is cool cos he doesn't have a lower jaw. Roxxors!!!!!

The game is definately easier to get into that AO, right click to attack, talk. Nicer GUI. Plus its Warcraft!!!!! I wanted to buy the BC today, but I'm going to put it off till tomorrow so I can test some more, then its off to the PvP server Frostmourne, my cousins are there and they're long time players so they can show me the ropes.

I would honestly quit AO right now except for the fact that I feel indebted to the guild and that we all can't play WoW since gotta pay after 10 days. So don't count me out of AO, mighty Jannorath will not abandon you. I am the Janitor of Wrath, mopping up mobs wherever they spawn, disinfecting temple floors Rubi-ka wide.

Now all I need is some bloody good armour!!!!! OH GOD WHY WON'T NO ONE MAKE CARBONUM!!!! I guess its time to take my request clanwide instead of just limiting it to the guild.

Good Boss helped me plant my tower. Its so nice to hit faster, I just got 20 levels worth of Melee Initiative ^_^ Now all I gotta do is get more tokens. You guys decide if you want to run missions or go Temple.

Don't panic.
What's up bitches. I know I kind of fell of the face of the planet for a while, but actually I was just pretending to be busy.

Rest assured, quite a lot of things have happened in my life, and some things that make you go THATSHITISBANANASMAN!!!

To illustrate the absurdity of the conundrums and lovable antics I get myself into, this shall be a big picture post, so big it makes you want to call your mother a whore to her face.

On some weekend on some day at some night time time, a dearest family friend known as Uncle Guna took Kavi, Divi, myself and his daughter Geetha to Damansara Club for dinner. He told us it was to celebrate Kavi's SAM results and my SPM results (LOLOLOLOLOLOXOXOXOXOO). It was very kind of him, and very very belated. But anyway, this is one kind, generous thoughtful person. No seriously. God couldn't create a better human if he shat on a saint.

Predictably, we took pictures, because a certain camera-crazy human BEAN was there, named Kavitha.

In other news, I've started my A-Levels at Brickfields Asia College. The subjects I'm taking are : Business Studies, Sociology and English Literature. Excluding the whole typical collegiate experience, I have to mention that my college's student body isn't very big, and its seriously as though all the students know each other regardless of what program they're in. Its kinda cool, but I joined college 3 weeks in the new term (their intake was in March, I joined in April, it was kind of a last minute decision) so everybody had made friends and already had their own groups. So ever since I've joined I've been hanging out with a lot of different people, mamaking with Business students and going to MidValley with A2 students (A Levels students a year senior to me. Meaning they finish their studies soon) after class. Its quite nice, and because this college doesn't have the 'combination system', I have different friends in every class too.

Also, classes for 2007 intake A Levels students on Monday have been cancelled, because the owner of the college has bought out an entire theatre hall in KLCC so we can all go watch Spiderman 3 at 11 am.

Can you say THISSHITISBANANASMAN??????!!!!

We got our tickets today during Sociology. Pretty cool. So yes, I am enjoying life as a college student. The only thing that sucks a little is my lectures are 3 hours long.

I'm watching you


Well my work day is ticking away and I'm running out of time to finish the shit I need to. On top of that I've just been informed that I have to go on a business trip to Bangkok next week for 2 days WITH the FAG, mentally unstable lunatic asshole in my department that I truly madly deeply want to drop dead. Although the process of dying should consume as much time as possible.

Sigh. Sigh!! SIGH!!!!!! Bloody shit. Oh well life goes on, at least I can get plastered while flying business class.

So anyway, I read in the shoutout box that Warlock (and most likely the rest) are coming down this week. Is it possible that we can resurrect out long long DotA sessions this Saturday morning? I know for a fact that Chino can't make it but if clown or newbie (with a total of at least 6/8 players) can come then we could have a session. Otherwise I'll be sitting I KL. I am curious if the thing that happened is intertent/TMnet related since there was serious lag yesterday.

Not much been happening over in KL lately. I've been bored aside from when I work and play games. Its difficult to do stuff with people here and co-workers are too old to go out and have fun (understandable but it still sucks for me).

We've become heavily addicted to Anarchy Online now, or at least I have which is why I am setting enforced limits to my playtime (hey I've got other games to play and pron to watch). Its loads of fun to play with people online and I've met and played with a ton of folks already. One thing that does suck a bit though in this game is that at times you can spend half to one whole hour just running around. AO really needs town portal scrolls/nanos.

I saw "Spiderman 3" and loved it. A fitting end to the series, but who knows there may be more. Loved the fact that they put Venom in the movie except for the part that he doesn't speak in plurals, no classic "We are Venom" lines. I mean ok maybe he didn't have to call himself Venom, but he still should have spoken in plurals. Now I'm just waiting for "Pirates of the Caribbean 3"

Lately I've been feeling extremely pissed off at the religious extremists in this country, and then became even more pissed off because it dawned on me that these are the regular folks. JAIS is a government body to maintain muslims etc. The Government funds the detention centres. The people vote for the Barisan Government time and time again. The people that complain never vote. My dad complains all the time and he has never vote his whole life!! Now I am guilty of this because I could go and register for the next election, you know make a difference. I have given up on Malaysia. I really have. I know it sounds weird to hear me say that since I've been hanging around for so long, but I think the country is heading down the toilet, although I do plan to enjoy what I can here while I'm here.

People don't see though, they never see, never bother until something horrible happens. I generally base my conclusions on religious extremism in Malaysia on one fact..extremists NEVER stop on their own. They keep going and going and getting worse and worse and by the time this so called silent majority of the 'beautiful religion' decide to act it will be too late, too entrenched, until even soundwaves will be able to remove it. How long do people have to do nothing to arrest a situation before they become culpable and resposible for the end result.

The Germans were responsible for Hitler and the Nazis, they could have stopped the problem earlier, but let it go on until they became Nazi Germany and lost many of their freedoms, but by then it was too late to do anything, the people in power could execute dissenters with ease.

People talk about racism in America, well racism exists everywhere because people are assholes. But at least white people don't get a 5-10% discount on houses over me. I could go on but enough has been said about this. If I ever become famous overseas I am so going to tell on everything here.

Well back to other things. My brother is coming back at the end of the months so at the very least I'll get a fair bit of outings there from chinese food dinners (mmmm sizzling japanese tofu) and DotA and probably a trip to Manila as well. At the very least if all the San Baka come down we can have DotA so all is happy. We should all go and see "Transformers" too, even if it sucks ass (I worry that it will but let's keep our fingers crossed).

I changed the comment format of the blog so that it won't open in a pop-up when we want to comments, I hope no one minds since this pop up thing really screws up the work PC IE and I can almost never comments. Warlock, feel free to change it back if this is a problem.

I've been having sinus problems again and its annoying since I spend a whole month repeatedly seeing a doctor to get it treated. I'm going to shoot for some traditional remedies now since I'm trying to avoid any kind of surgery.

Well I've got to be off to finish a document so that I can go home and log into AO to plant my tower and get so much needed melee initiative (god the hammer hits so slowly)

This is the 600th post....uhm.......well c.........the this number is too great to resist, i just have to post something.....

uhm...ok, I will try to get AO asap, my connection is so fucked up and i reckon complaining will do nothing good and it wont solve it, afterall, it wont be like this now if they really solve it years back. Still hanging on the downloading now.

I will also be trying out Granado Espada when the open beta starts. There is another game to play, secondlife, i dunno much about this game, feel free to visit www.secondlife.com my best gueses is that this is not an action game, erm...obviously. so, i think Kavi might be interested and i myself is interested, but too bad, my hdd space have limit. >.< will play all the game if i can. Ok, got some time for a quick post. Anarchy Online rocks !

So far these are the characters we have and are actively running in a party :

Adventurer - Human - Swords
Enforcer - Atrox - 2 handed blunt weapon
Agent - Optifex - Rifle
Martial Artist - Optifex - bare hands

(yes , besides the Agent , everyone else is very 'kampung' with traditional weapon .. even though the game is set in the future :p)

It might be better for anyone else to try out other characters but if you got your heart set on the same characters it's not really a problem.

I recommend that for the sake of speeding up the game, looting is open for all party members but I recommend that you open a custom window to display only all loot items garnered so you can just request for anything that you want. The problem will come when two people specialise in the same weapon or items but that can be settled between them ..

When in the Newbie Island where you start a new character, you will be in an instance out of a few ones so you might not see your friends even if they are on the same island. To meet up you have to go into the teleport tower (non-instanced) party and then go out. Once you leave Noob island you cant come back ..

I recommend not wasting time in Noob Island, just finish all quest then get all the Noob Armour (must !) then get shades from the vendor (if you want) then when you are lvl 8-10 just leave and look for us to help you power level :p Dont bother stacking up the medi-kits and nana-regens by doing the basic missions (it is crap .. i stacked 1000 + for each .. went out of Noob island and saw what they were selling and just threw mine away)

We are all in the CLAN faction . If you join Omni-Tech we can not party .. of course we can meet up , then we kill you and get some PvP points :p

p.s : look for something called clicksaver .. not sure wat it is but it's helpful in getting the right missions for you
